For some outcomes, yes—but the answer depends heavily on what you expect it to do.

Caffeine + L-Theanine: Does the Combination Work?
The combination of caffeine and L-theanine has more controlled human research than most commercial “nootropic stacks.”
A 2025 systematic review and meta-analysis included 50 randomized controlled trials investigating tea, L-theanine, or L-theanine plus caffeine in healthy participants. Fifteen trials contributed data to at least one meta-analysis.
For L-theanine plus caffeine, pooled findings favored the combination over placebo for selected cognitive measures during the first two hours after intake.
For example, significant differences were reported for digit-vigilance accuracy during the second hour and attention-switching accuracy during the second hour.
Choice reaction time during the first hour also favored the combination numerically, although its confidence interval crossed the null.
There is no scientifically established 2:1 “golden ratio.” Human studies have used multiple combinations of caffeine and L-theanine. A popular 200 mg L-theanine / 100 mg caffeine combination should not be presented as universally optimal.
Older trials and systematic reviews also support potential benefits for selected attention measures from caffeine plus L-theanine.
But L-theanine does not neutralize caffeine.
The combination cannot be assumed to eliminate caffeine-related sleep disruption, nervousness, palpitations or other adverse effects in susceptible individuals.
Does L-Theanine Help Sleep?

The pooled standardized mean differences were modest for some outcomes: approximately 0.15 for subjective sleep onset latency, 0.33 for daytime dysfunction and 0.43 for the overall subjective sleep-quality score.
The authors specifically highlighted the limited number of studies investigating pure L-theanine.
They concluded that additional research is needed to determine an appropriate dose and duration for improving and maintaining sleep quality.
Best interpretation: L-theanine may support selected sleep outcomes, particularly subjective sleep measures. Current evidence does not establish it as a treatment for clinical insomnia or prove that one bedtime dose works for everyone.
A second systematic review was published in 2026 in Nutritional Neuroscience after appearing online in 2025.
That review identified 13 standalone L-theanine trials involving 550 participants, including 11 randomized controlled trials and two single-arm open-label trials. Doses ranged from 50 to 900 mg/day.
