SMALL HUMAN TRIALA randomized double-blind placebo-controlled study published in 2009 enrolled 30 Japanese adults aged 50–80 with mild cognitive impairment.
Participants receiving Lion’s Mane dry powder for 16 weeks improved on the study’s cognitive scale compared with placebo at several time points.
However, the study was small, involved people who already had mild cognitive impairment rather than healthy adults, and cognitive scores declined again after supplementation stopped.
Healthy Younger Adults
MIXED / PRELIMINARYA 2023 randomized pilot trial studied 41 healthy adults aged 18–45 using 1.8 g of Lion’s Mane for 28 days.
Researchers observed faster performance on one Stroop outcome following a single dose and a statistical trend toward reduced subjective stress following chronic supplementation.
Importantly, the researchers also reported null and limited negative findings and emphasized the small sample size.
A separate randomized crossover study published in 2025 examined an acute standardized Lion’s Mane fruiting-body extract in 18 healthy younger adults. It found no significant overall improvement in composite cognitive performance or mood, although one individual motor-performance test improved.
Mood, Anxiety & Stress
LIMITEDA small 2010 randomized study included 30 women who consumed Lion’s Mane-containing cookies or placebo for four weeks.
Some questionnaire outcomes improved, but the study was small and several reported findings were within-group changes or trends rather than robust evidence of a broad antidepressant or anti-anxiety effect.
This is enough to justify further research—not enough to call Lion’s Mane an established treatment for depression, anxiety or stress.
